Eagle MEM ①是根据 Harry Eagle 1959 年开发的最低必需培养基(MEM)配方生产,用于细胞培养。一般这类培养基无法进行高温高压灭菌,因为会使其成分降解或改变。岛津 Eagle MEM ①培养基采用特殊工艺,可进行高温高压灭菌而不会破坏其中的成分。该培养基对 Hela 细胞、L 细胞和其他细胞系以及从生物组织中分离出来的未建立的细胞具有良好的促生长作用。
称取 9.4g 该粉末培养基溶解在蒸馏水中,制成 1000mL 溶液,然后在 121℃条件下高温高压灭菌 15 分钟。之后将已经过滤灭菌或在密封状态下进行高温高压灭菌的 10% 碳酸氢钠水溶液适量加入冷却至室温的培养基中(添加 12.5-22.0mL,5% CO₂ 气体下,37℃的 pH 值为 7.1-7.4)。如需长时间保存,请密封保存在阴凉干燥处(2-10℃)。使用前,添加已经过滤灭菌的谷氨酰胺 0.292 克,根据需要添加适量的血清。
