Long Term Storage: +4°C
Use/Stability: 20S Proteasome remains relatively stable and active so long as repetitive freeze-thaw cycles are avoided. It has been demonstrated that 20S proteasome undergoes only slow loss of activity when stored at 4ºC for up to six months and longer. The material is supplied at ~4ºC. If it is to be used on a regular basis, it is recommended that the material should be stored at >0ºC-4ºC, however, freezing must be avoided. For longer term storage the material may be aliquoted into single use aliquots and stored at –80ºC with the addition of up to 50% glycerol (high purity). If BML-PW8720 is stored frozen then it must be used immediately upon thawing as experimentation has shown that, whilst activity may be retained (or even enhanced) during freezing, activities may decrease very rapidly after thawing. The reason for this potential post-freeze-thaw instability is poorly understood.
Handling: Avoid freeze/thaw cycles.
Miscellaneous/General: The 20S proteasome is the catalytic core of the 26S proteasome, the central protease of the ubiquitin pathway of protein degradation. Unlike the 26S proteasome, which degrades proteins in an ATP-dependent manner, the 20S proteasome is ATP-independent and only degrades entirely unfolded polypeptides.
