英文名: ZLN024

描述: ZLN024 是有效的 AMPK 变构激活剂，直接激活重组 AMPK α1β1γ1，AMPK α2β1γ1，AMPK α1β2γ1 和 AMPK α2β2γ1 异源三聚体，EC50分别为 0.42 μM，0.95 μM，1.1 μM 和 0.13 μM。

体外活性: ZLN024 通过变构增强活跃的AMPK异三聚体以及非活性的α1亚单位截断体α1 (1-394) 和 α1 (1-335)，但不包括α1 (1-312)。对于α1β1γ1，ZLN024将活性提高了1.5倍，其EC50为0.42 μM；对于α2β1γ1，活性提高了1.7倍，EC50为0.95 μM。此外，ZLN024直接激活重组的AMPK α1β2γ1，活性提升1.7倍，EC50为1.1 μM；对AMPK α2β2γ1，则提高了1.6倍，EC50为0.13 μM。

体内活性: C57BKS db/db mice are administered a 15 mg/kg/day dose of ZLN024 by daily gavage for 5 weeks; 250 mg/kg/day Metformin (Met) is used as a positive control. After 4 weeks of treatment, ZLN024 improves glucose tolerance. ZLN024 reduces the fasting blood glucose by 15%. Liver tissue weight, triacylglycerol and the total cholesterol content are decreased.
