英文名: Elacestrant

描述: Elacestrant (RAD1901) 是一种具有口服活性的、选择性的雌激素受体 (ER) 降解剂 (SERD)，能够作用于 ERα (IC50:48 nM)和 ERβ (IC50:870 nM)。

体外活性: Elacestrant的治疗表现为剂量依赖性地抑制ERα表达，其EC50为0.6 nM。对ER阳性的MCF-7细胞用E2处理会引起强效的剂量依赖性增殖，EC50为4 pM。在10 pM E2存在的条件下对细胞进行Elacestrant治疗，会出现剂量依赖性的增殖减少，IC50值为4.2 nM。Elacestrant特异性结合并降解ER，并且是ER阳性乳腺癌细胞增殖的强效拮抗剂。

体内活性: Elacestrant在MCF-7异种移植模型中显著有效地抑制了肿瘤生长。此化合物能够保护卵巢切除引起的骨丢失，并且阻止了E2的子宫增生效应。接受Elacestrant治疗的动物比对照组或接受fulvestrant治疗的动物存活时间更长。
