描述: 4-Aminohippuric Acid 是诊断剂，能够用于肾脏检测，在肾血浆流量的测定中使用。

体外活性: 4-Aminohippuric acid在正常个体中的肾提取比率约为0.92.提取后,大部分4-Aminohippuric acid通过近端小管主动分泌,并完全通过肾脏排出.4-Aminohippuric acid 的清除力用来衡量肾血浆流量.4-Aminohippuric acid也用来测量肾小管分泌机制或转运极限.已经通过在大鼠肾脏中停止流动毛细血管灌注方法测量了4-Aminohippuric acid摄入皮质肾小管细胞的动力学.在大鼠肾衰竭模型中,与正常大鼠（29.30mL/min/kg）相比,尾静脉注射3.0 mg/kg Uranyl nitrate引起CLPAH显著降低（0.89mL/min/kg）.慢性肾功能衰竭的大鼠显示药代动力学发生改变,且肾脏排泄4-Aminohippuric acid.与Sham大鼠 (μL/min/g,145 vs. 100,39 vs. 20和108 vs. 70,)相比,慢性肾功能衰竭大鼠4-Aminohippuric acid排泄、过滤和分泌的负载均减弱,匀浆和膜中表达的OAT1减少约40％.因此,通常测量4-Aminohippuric acid的肾脏消除以反映肾脏的有机阴离子转运能力.

体内活性: 10 mmol/L 4-Aminohippuric acid通过表达MRP2的HEK293细胞的膜囊泡,转运率为 21.9 pmol/ mg protein/ min。4-Aminohippuric acid的Km值为880 mM,Vmax值为2.3 nmol/mg 蛋白/ min。4-Aminohippuric acid通过上皮细胞表达的有机阴离子转运体而进行转运。 通过在HEK293细胞上转染并表达的NPT1摄取4-Aminohippuric acid是可饱和的并遵循Michaelis-Menten动力学,具有接近2.66 mM的Km和940 pmol/mg蛋白质/ 30s的Vmax值。4-Aminohippuric acid也通过OAT以外的转运体进行转运,包括NPT1和MRP2。
