英文名: Resveratrol
描述: Resveratrol (SRT 501) 属于多酚类天然产物,是一种植物抗毒素,具有抗氧化和化学预防活性。Resveratrol 的靶点广泛,包括 COX、SIRT、LOC 等。Resveratrol 可以诱导细胞自噬和凋亡。
细胞实验: HAECs were treated with RSV in the presence of isobutylmethylxanthine (IBMX). The reaction was stopped by adding ice-cold 6% trichloroacetic acid and the supernatant fractions of the cellular lysates were extracted, dried and acetylated. Cyclic GMP levels were determined by an enzyme immunoassay kit and results standardized by protein levels [5].
动物实验: Eight-week male apoE-/- mice (C57BL/6 background) were used in the study. Mice were housed in photobiologic light-exposure chambers with a 12:12-h light:dark cycle, and eat food ad libitum. Mice were randomly divided into two groups: one group receiving a high cholesterol diet (HCD, 21% fat, 19.5% casein, and 1.5% cholesterol), the other receiving HCD supplemented with RSV (200 mg/100 g diet). All mice were fed for 8 weeks. To identify the potential role of PKA, some mice were treated with PKA specific inhibitor (KT5720, 1.2 mg/kg) via intraperitoneal injection once a day at the last 4 weeks. The animal experiments were conducted according to the institutional guidelines of Guangdong Provincial People's Hospital [5].
体外活性: 方法:小鼠乳腺癌细胞 4T1 用 Resveratrol (50-250 μM) 处理 72 h,使用 CCK-8 检测细胞活力。结果:Resveratrol 剂量依赖性抑制 4T1 细胞的活力,IC50 为 93 μM。[1]方法:人乳腺癌细胞 MCF-7 用 Resveratrol (1-100 μM) 处理 24 h,使用 luciferin-luciferase assay 检测细胞内 ATP 含量。结果:Resveratrol 降低了细胞 ATP 含量。在 1 μM Resveratrol 时降低了约 16%,在 100 μM Resveratrol 时降低了 50%。[2]
体内活性: 方法:为检测体内抗肿瘤活性,将 Resveratrol (50-100 mg/kg,5% ethanol and 25% polyethyleneglycol 400 in distilled water) 腹腔注射给携带人卵巢畸胎瘤 PA-1 的 BALB/c (nu/nu) 小鼠,每天一次,持续四周。结果:Resveratrol 抑制 PA-1 细胞异种移植物的生长,抑制 eEF1A2 的表达。[3]方法:为研究对抑郁症小鼠的影响,将 Resveratrol (10-30 mg/kg) 腹腔注射给抑郁症模型的 C57BL/6 小鼠,每天一次,持续二十一天。结果:Resveratrol 可显著提高前额叶皮层神经递质 DA 和 5-HT 的水平,并增加大脑中 NPY 的表达,这对抑郁症起到拮抗作用。[4]
