英文名: Prucalopride

描述: Prucalopride (R-93877) 是一种选择性、高亲和力的 5-HT4A/4B 受体激动剂，Ki 为2.5和 8 nM。它对 5-HT4A/4B 受体的选择性是其他 5-HT 受体亚型的 290 倍以上。

体外活性: Prucalopride（2/4 mg）给药得到的每周完整排便分别为30.9%和28.4%,而安慰剂对照组为12.0%.接受2 mg Prucalopride的患者（47.3%）和接受4 mg Prucalopride的患者（46.6%）会增加自发性的次数,平均每周完整的排便为一次或更多,而安慰剂对照组为25.8%.Prucalopride（2/4 mg）使其它次要疗效终点均显著提高,包括患者对他们肠功能和治疗的满意度及对便秘症状严重度的感觉. Prucalopride（4 mg/day）使便秘患者整体的胃排空和小肠转运加速,但不引起肠道排空障碍.Prucalopride（4 mg/day）可使整体的结肠运输加速,并增加结肠的排空速度.

体内活性: Prucalopride（1 mM）可使电刺激后豚鼠近端结肠的反弹收缩被显著放大。Prucalopride可浓度依赖性地诱导收缩（pEC50：7.5）。Prucalopride对大鼠食道粘膜肌层的放松有诱导作用（pEC50：7.8）,出现单相浓度响应曲线。
