英文名: Macitentan

描述: Macitentan (ACT-064992) 是一种内皮素受体拮抗剂，用于治疗肺动脉高压。

体外活性: 在高血压DOCA盐鼠中,Macitentan（10 mg / kg）以剂量依赖的方式降低平均动脉血压. 在肺动脉高压野百合碱大鼠模型中,口服给药Macitentan（30 mg / kg /天）剂量依赖性地预防肺动脉高血压的发展和右心室肥大的发展. 在2型糖尿病诱导db / db小鼠中,口服Macitentan（25 mg / kg / day）,能够降低mRNA表达,同时增加NF-κB的激活.

体内活性: 在原代人类肺平滑肌细胞中（IC50=1 nM）,Macitentan抑制ET-1诱导的细胞内钙离子增加。在离体的大鼠主动脉环中,Macitentan抑制ET-1诱导的收缩。在离体的大鼠气管环中,Macitentan抑制S6c诱导的收缩。
